Hengrui Medicine's GIP/GLP-1 Dual-Target Drug Ruipupotide NDA Accepted by China's NMPA
nashnova research
Hengrui Medicine's self-developed GIP/GLP-1 dual-agonist ruipupotide has entered China's regulatory review, backed by two Phase III trials — this means Eli Lilly's tirzepatide is about to face its first domestic challenger in the world's largest diabetes market.
What is this drug and what does it do?
Ruipupotide is a GIP/GLP-1 dual-target agonist — a single injection that activates two insulin-promoting pathways at once — filed for adult type 2 diabetes (T2DM).
In plain terms = it flips two "blood-sugar switches" simultaneously: one boosts insulin secretion, the other improves the body's response to insulin, while also suppressing appetite and regulating lipid metabolism.
Developed in-house by Hengrui's subsidiary Fujian Shengdi, with cumulative R&D spending of roughly RMB 746 million (unaudited).
Is the clinical data strong enough?
The filing rests on PRIME-1 and PRIME-2, two pivotal Phase III trials enrolling a combined 1,102 patients across 147 sites in China.
PRIME-1 (vs. placebo): the ruipupotide 4 mg arm cut HbA1c — a marker of average blood sugar over three months — by 2.67% at week 36.
PRIME-2 (vs. semaglutide 1 mg): the ruipupotide 4 mg arm cut HbA1c by 2.78%, meeting non-inferiority — and on the key secondary endpoint, it was statistically superior to semaglutide (one-sided p<0.0001).
This means → in a head-to-head trial, the high-dose ruipupotide arm beat the most widely used GLP-1 mono-agonist on glycemic control.
Whose market is it challenging?
Only one GIP/GLP-1 dual-agonist is approved globally: Eli Lilly's tirzepatide (brand name Mounjaro), launched in the US in 2022 and approved in China in May 2024.
Mounjaro posted roughly $22.97 billion in global sales in 2025, making it one of the top-grossing diabetes/obesity drugs worldwide.
This means → if approved, ruipupotide would become China's first domestic GIP/GLP-1 dual-agonist, entering a high-growth lane Lilly currently owns alone.
How big is the Chinese market?
Global diabetes patients reached 589 million in 2024; China alone accounted for roughly 148 million, the largest national population.
T2DM makes up over 90% of all diabetes cases — exactly the indication ruipupotide has filed for.
In plain terms = China holds the world's largest pool of potential patients, and a competitive domestic dual-agonist has a massive addressable market by default.
What to watch next?
NDA acceptance is only the first step into the review pipeline — whether and when approval comes remains uncertain.
Even with approval, ruipupotide must overcome tirzepatide's established prescriber habits and brand awareness — the commercialization challenge for a late mover can rival the clinical hurdle.
On safety, the company says tolerability is consistent with other GLP-1-class drugs, but real-world post-marketing data will be the definitive test.
